Conventional silica-based RNA purification kits suffer from size and GC bias
Conventional RNA purification kits isolate RNA using a simple silica-based matrix. This results in a biased RNA profile, with smaller RNAs (< 200 nt) missing. In addition, there is often a significant bias against GC-rich sequences. The image on the right shows a comparison of total RNA isolated using either a Norgen Total RNA Purification kit, or a kit from competitor Q. The loss of RNAs smaller than 200 nt in size with competitor Q's kit is clearly visible.
