Ultrafast live F-actin imaging with SPY FastAct probes

SPY FastAct™ probes are new and unique fluorescent live cell actin probes. They label very dynamic actin filaments as well as the more static ones. SPY650-FastAct™, SPY555-FastAct™ and SPY700-FastAct™ are bright, fluorogenic, non toxic, cell permeable and highly specific live cell probes for fluorescence imaging of actin. They don't require any transfection and work just by adding the probe to the cells and performing imaging a couple of hours later.

How does SPY650-FastAct work?

SPY650-FastAct™ is a bright, far red, fluorogenic & nontoxic F-actin stain based on our SPY™ dyes series. Its optimized structure allows to label F-actin in live cells with high specificity and low background. The unique and unmatched feature of SPY650-FastAct™ is its ability to label very fast actin dynamics, like LifeAct™ but the probe does not require any genetic manipulation, transfection or overexpression of fluorescent proteins. SPY650-FastAct™ is based on our bright, photostable & far red SPY650 fluorophore which is far superior to fluorescent proteins.

SPY650-FastAct™ enables multicolor imaging with SPY505, SPY555, SPY595, SPY700, GFP or m-cherry. SPY650-FastAct™ can be imaged with standard Cy5 filterset. It can be used for widefield, confocal, SIM or STED imaging in living or fixed cells and tissue.

SPY555-FastAct: a near-red alternative to SPY650-FastAct

SPY555-FastAct™ is a bright, near red, fluorogenic & nontoxic F-actin stain based on our SPY™ dyes series. Its optimized structure allows to label F-actin in live cells with high specificity and low background. The unique and unmatched feature of SPY555-FastAct™ is its ability to label very fast actin dynamics, like LifeAct™ but the probe does not require any genetic manipulation, transfection or overexpression of fluorescent proteins. SPY555-FastAct™ is based on our bright, photostable & near red SPY555 fluorophore.

SPY555-FastAct™ enables multicolor imaging with SPY505, SPY595, SPY700, GFP or m-cherry. SPY555-FastAct™ can be imaged with standard TMR/Cy3 filterset. It can be used for widefield, confocal, SIM or STED imaging in living or fixed cells and tissue.

SPY700-FastAct™

SPY700-FastAct™ is a new and unique fluorescent live cell actin probe. It labels very dynamic actin filaments as well as the more static ones. SPY700-FastAct™ is a bright, fluorogenic, non toxic, cell permeable and highly specific live cell probe for fluorescence imaging of actin. It does not require any transfection and works just by adding the probe to the cells and perform imaging a couple of hours later.

Image and movie by Maxime Cammeraat, from Margadant lab, Amsterdam, NL. 2 populations of primary human endothelial cells were separately labeled with SPY595-DNA or  SPY505-DNA. After the DNA labeling, the 2 populations were trypsinized, mixed together,  stained with with SPY700-FastAct and imaged.

SPY650-FastAct_X and SPY555-FastAct_X: probes for dynamic and static actin

SPY650-FastAct_X and SPY555-FastAct_X are our most advanced live cell actin imaging probes. They label both highly dynamic and static actin filaments and are bright, fluorogenic, non toxic, cell permeable and highly specific. Moreover, they completely lack undesired F-actin stabilization and toxic side effects. They do not require any transfection and work just by adding the probes to the cells and performing imaging a couple of hours later.

Ordering information

1 vial of SPY650-FastAct™, or SPY555-FastAct™ or, SPY700-FastAct™ in lyophilised form allows preparation of 50 ul of 1000x DMSO stock solution, sufficient for 100 stainings.
