Broad researchers describe new CRISPR-based method called Prime Editing
The goup of David Liu at the Broad Institute has published a paper in Nature describing a novel approach to edit genomes. The method, called "prime editing" relies on a fusion between a catalytically impaired Cas9 enzyme and a reverse transcriptase. Prime editing directly writes genetic information into the genome using just a single guide RNA. No double strand breaks or exogenous DNA templates are needed for the modification.
CRISPR enters first human clinical trials
Since its debut, CRISPR gene editing has held the promise of curing genetic diseases. Now - seven years after the first CRISPR publication - the first clinical trials using the gene scissors are starting.
Clinical trials now beginning include the use of CRISPR/Cas9 to combat cancer and blood disorders. The strategy is to remove some of a person’s cells, edit the DNA and then inject the cells back in, now hopefully armed to fight disease. Researchers are also designing clinical trials to treat people with an inherited blindness. Those trials, if successful, could spur future trials for Duchenne muscular dystrophy, cystic fibrosis and a wide variety of other genetic diseases, affecting millions of people worldwide.

CRISPR DNA base editors cause tens of thousands of off-target RNA mutations
A team of Chinese researchers has found that CRISPR-based DNA base editors can induce tens of thousands of off-target mutations in RNA. Adenine base editors (ABEs) and cytosine base editors (CBEs) are known to efficiently and precisely edit point mutations in DNA with minimal off-target DNA editing. However, recent studies have also shown that despite their reputations for generating low levels of off-target DNA edits, base editors may create unexpected problems.

A novel approach for targeted genomic insertion of DNA fragments
Researchers from the National Institutes of Health and the Broad Institute report a novel system for precisely inserting DNA into a genome that uses a combination of a CRISPR-associated transposase (CAST) and a CRISPR effector protein (Cas12k) from the cyanobacterium Scytonema hofmanni.
Their system, termed "ShCAST" catalyzes RNA-guided DNA transposition by unidirectionally inserting segments of DNA 60 to 66 bp downstream of the protospacer site with high efficiency. Inserting DNA fragments into the E. coli genome via ShCAST yielded insertion efficiencies of up to 80 percent, without the need for positive selection.

The introduction of the CRISPR-Cas9 system for genome editing has been an incredible boon for biological research. As our understanding of CRISPR technology increases, so does our awareness of the off-target editing events carried out by Cas9 nuclease. Existing CRISPR guide RNA design tools, such as CRISPR Design (MIT [1]) and CCTop (CRISPR-Cas9 Target online predictor, Heidelberg University [2]), attempt to predict off-target editing of the guide RNAs. However, these tools are limited by an incomplete understanding of the factors that cause Cas9 to cleave at incorrect sites.
