Abstract:
CRISPR-Cas9 has been widely employed for a variety of genome engineering applications. However, Cas9 can bind and cleave partially complementary off-target sequences, which raises safety concerns for its use in clinical applications. In this webinar, we will discuss the recent structural investigations that shed light on the molecular mechanisms underlying Cas9 specificity and off-target discrimination. We also highlight how these experimental results can help to develop a more efficient and specific CRISPR-Cas9 gene editing system.
